Το 1969 συνεργάστηκε με τους The Beatles για τα λεγόμενα “Get Back sessions” και το άλμπουμ “Abbey Road”. Ο John Lennon είχε προτείνει μάλιστα την ιδέα, ο Preston να ενσωματωθεί στο συγκρότημα σαν ο “Fifth Beatle”. Το 1972 γνώρισε επιτυχία με το ορχηστικό θέμα “Outa-Space”, το οποίο ανέβηκε μέχρι την δεύτερη θέση του Billboard Hot 100 και του χάρισε το βραβείο Grammy “Best Pop Instrumental Performance”. Το 1978 εμφανίστηκε στο φιλμ “Sgt. Pepper’s Lonely Hearts Club Band” του σκηνοθέτη Robert Stigwood. Θεωρείται μια από τις σημαντικότερες μουσικές προσωπικότητες του 20ου αιώνα.
